Emerging role of Elovl5 in central and peripheral nervous system

ELOVL5 is an enzyme that elongates long chain fatty acids, with a marked preference for polyunsaturated molecules. Mutations of ELOVL5 cause the spino-cerebellar ataxia type 38, indicating an important role in the central nervous system. Starting from this pathological point of view, we are trying to better understand its localization and role in CNS and PNS. In particular, we are focusing on its involvment in myelin formation and correct action potential conduction along the axon. 

Moreover, Elovl5 enzyme is mainly expressed in Purkinje cells and it has a crucial role for the synthesis of AA and DHA, two PUFAs that have varied functions in the brain. For this reason, its lack might have a negative impact on neuronal function, in particular on cerebellar circuitry signalling and short- and long-term synaptic plasticity in the cerebellum.
